Blunt ended cDNA libraries for 454 sequencing provide an innovative, yet cost-effective and efficient approach to gene discovery and annotation. 454 sequencing combined with laser-capture microdissection (LCM) and polymerase chain reaction (PCR) enables the isolation, massive cloning, and subsequent sequencing of transcripts from specific cell types. Helica Biosystems Inc. now offers blunt ended cDNA libraries containing specific transcripts that can be anchored to gene-specific regions in the 3' untranslated region of a cDNA. Anchoring the specific transcripts to distinct polymorphisms in the 3' untranslated region allows for the simultaneous sequencing of large, informative base sequences with the synthesis of the complementary strand. Blunt ended cDNA libraries reduce the number of reads necessary to identify distinct mRNAs without using complete cDNA libraries.
